Eight weeks of daily supplementation with 30 billion CFU of Pediococcus acidilactici PA53 in 75 adults aged 55–70 produced measurable improvements across multiple domains: happiness scores rose 12.7% (MUNSH scale, P<0.05), sleep quality improved 23% (PSQI, P<0.05), and gastrointestinal symptoms declined 9.5% (P<0.001). Inflammatory markers dropped substantially — serum IL-6 fell from 135.6 to 108.4 pg/mL and TNF-α from 65.8 to 54.2 pg/mL — while the gut-barrier cytokine IL-25 rose 40%. Microbiome shifts included near-doubling of Ruminococcus, and meaningful gains in Faecalibacterium (6.9%→10.6%) and Blautia (7.6%→11.4%), all genera associated with short-chain fatty acid production and anti-inflammatory signaling.
This trial is notable for linking a single probiotic strain to simultaneous emotional, inflammatory, and microbiome endpoints in a genuinely underrepresented demographic — healthy older adults rather than clinical patients. The gut-brain axis framework here is plausible: Faecalibacterium prausnitzii depletion correlates with depression in prior cohort studies, and the IL-6 reduction aligns with known inflammaging pathways that accelerate cognitive and mood decline after 60. That said, the study is small (75 participants, ~37 per arm), confined to one ethnicity, and eight weeks is insufficient to assess durability. Perceived stress improvement failed within-group significance, tempering the emotional health narrative. The finding is best categorized as confirmatory and mechanistically suggestive — it strengthens the case for strain-specific probiotic trials in aging populations but falls short of practice-changing evidence without longer, larger replication.
