For adults navigating an overwhelming supplement marketplace, the challenge has never been distinguishing hype from evidence — it's identifying which compounds actually engage the biological machinery of aging itself. A new systematic review does exactly that, mapping specific nutrients to the molecular pathways most implicated in age-related decline: chronic low-grade inflammation, mitochondrial deterioration, and progressive loss of muscle and metabolic resilience.

Drawing on 32 human studies sourced from PubMed, Scopus, and Web of Science, the review evaluated nutritional compounds across five functional domains: mitochondrial health, cognitive performance, immune regulation, metabolic function, and musculoskeletal maintenance. Among the strongest signals: protein and collagen supplementation, particularly when combined with resistance training, produced consistent improvements in muscle mass and physical function in older cohorts. NAD⁺ precursors — notably nicotinamide riboside and nicotinamide mononucleotide — along with antioxidants, glycine, and N-acetylcysteine demonstrated measurable effects on mitochondrial efficiency and cellular stress resilience, though effect sizes varied substantially depending on population characteristics and dosing protocols. Probiotics showed emerging relevance for immune and metabolic outcomes, though evidence here remains more preliminary.

This review sits at a meaningful intersection: it is neither a single compound trial nor a broad nutritional survey, but a mechanism-first mapping exercise. That framing matters because aging biology doesn't fail uniformly — mitochondrial decline may precede inflammatory dysregulation in some individuals, while sarcopenia dominates others. The honest limitation the review surfaces is consequential: heterogeneity in study designs, formulations, and biomarker endpoints makes population-wide dosing recommendations premature. Most included studies were short-duration and modestly sized, which limits causal inference. Still, the convergence of evidence around NAD⁺ biology and protein-exercise synergy represents more than incremental confirmation — it reflects a maturing understanding that supplementation works best as a complement to, not substitute for, foundational lifestyle behaviors.
